Playing League of Legend as well as World of Tanks, I find that in random games, there is very, very little prematch communication. While players can naturally fall into their roles, I think it would be to the game's advantage to have a pregame lobby where strategy, at least overall strategy, can be discussed without second by second time pressure. This will allow lancemates and companies to tune loadouts, discuss the map, and even trade tips and pair off.
This lack of interaction is particularly frustrating in LoL, where two players square off waiting for the other to pick so they can comp properly. And the 45 second timer is not long enough to change a mastery page for a champ you really aren't prepared for but end up playing to balance your team. Not saying you should have ten minutes to **** around in the mechlab, but the ability to lobby with a random team with a map queued up, and ready up before you enter the actual game queue, I think would add a good dimension to the game.
Unless, of course, the majority of players are "mash ready, run forwards, shoot."

Keep in mind that if you have a lobby that doesn't start the game till everyone presses ready, you might end up with a troll somewhere just never pressing ready and screwing with the whole lobby. (Unfortunately that does happen in a lot of PC games I have played in the past)

I like how they countered this in Mass Effect 3 with the ability to boot players who take too long. Unfortunately, this feature has it's own drawbacks as well.

Well, there could be a max timer, so if someone hasn't clicked "ready" within a certain amount of time, then the match starts automatically.

I've played game with both systems. I think that it should be about a 1 and 1/2 to 2 minute timer. That gives enough time to co-ordinate, but keeps the pace of the game as well.

If 2/3 of the players in a lobby have set themself to ready a timer starts to count down until the battle begins if the others be ready or not.
