As the title says what is the best time of the day to do 12 mans through out the week (Eastern Standard Time)

Anytime after 8:30-9pm est you can get groups but after 10 is the best time. ThAts when us east coasters are good to go and the west coasters are home from work.

The majority of team practices start at 9pm EST.

Most of the european clans play around 20:00h / 8:00 pm CEST (Central European Summer Time).
CEST is 2 hours ahead of Coordinated Universal Time (UTC) -> CEST = UTC + 2h.
The HanseRaiders are practicing Sunday, Wednesday and Thursday (around 8pm CEST) with one of their companies (and all other days with several random drops).

I could be wrong, but private 12-man matches may pretty much eliminate the 12-man pub queue.

Like Aduivo said, 9pm to 12 pm is usually an easy time to find a hit. Beware, if you are dropping on all modes you will probably find a team playing comp matches. They will ask you to not drop in that mode so they have an easier time syncing. It is good form in the 12 man queue to try to accommodate anyone asking.

Shouldn't be a problem after tomorrow's patch! No more need to sync drop the 12's.
